Overview

About Semax

Semax is the parent compound of the series: the ACTH(4-7) tetrapeptide MEHF extended at the C-terminus with Pro-Gly-Pro. That extension is a protease block — proline-rich C-termini resist carboxypeptidase cleavage — and it is what made a fragment with a half-life of seconds usable at all.

It is stocked alongside the terminally capped analogue because comparing the two is how the effect of terminal protection is measured. Where a protocol specifies Semax, the unmodified peptide is what it means; substituting the capped analogue changes mass, retention time and solubility and will not reproduce the original method.

Neurotrophin expression

The core reported effect of the series: upregulation of BDNF and NGF transcripts and protein in neuronal preparations.

Analogue comparison

Held alongside N-Acetyl Semax Amidate to isolate the effect of terminal capping on half-life and distribution.

Ischaemia models

Handling

Reconstitution and stability

Reconstitution

Reconstitute with sterile or bacteriostatic water added slowly against the inner wall of the vial. Allow the cake to dissolve without agitation, swirling gently if required. Do not vortex or shake.

Stability

Lyophilised at −20 °C, desiccated and protected from light, stable for at least 24 months. Reconstituted and held at 2–8 °C, use within 28 days. Avoid repeated freeze-thaw.

Notes

  • Bring the vial to room temperature before opening so atmospheric moisture does not condense onto cold powder.
  • Record the reconstitution date on the vial; concentration drift between runs is usually evaporation, not synthesis variance.
  • Aliquot at first reconstitution where the working programme needs more than a few withdrawals.

Should I use this or the amidate?
Use whichever your protocol specifies. They are different compounds. If you are designing new work, the capped analogue has the longer solution half-life; if you are reproducing published work, match what the original used.
Does it have corticotropic activity?
No. The ACTH(4-7) fragment lacks the residues needed for melanocortin-2 receptor activation.
Is it oxidation-sensitive?
Yes, mildly. The single methionine oxidises on air exposure in solution. Expect a small early-eluting peak in an ageing preparation.
