Overview

About Humanin

Humanin was the first mitochondrial-derived peptide identified, encoded within the 16S rRNA region of mitochondrial DNA. Like MOTS-c it demonstrated that mtDNA carries small open reading frames producing bioactive peptides, which reframed how the field thinks about mitochondrial signalling.

Handling is the practical challenge. The sequence contains a run of leucines that makes the peptide markedly hydrophobic and prone to aggregation in aqueous buffer. Most protocols prepare a DMSO stock and dilute into aqueous medium immediately before use; preparing directly into water frequently produces a suspension rather than a solution and a concentration that reads low.

Cytoprotection

The founding work identified humanin in a screen for protection against amyloid-beta toxicity, and cytoprotection remains its principal research use.

Mitochondrial signalling

Alongside MOTS-c, humanin is central to the study of mitochondrial-derived peptides as an outward signalling system.

Receptor pharmacology

Signalling through FPRL1 and gp130-family receptors is studied to establish how a mitochondrially encoded peptide reaches cell-surface receptors.

Reconstitution and stability

Reconstitution

Reconstitute with sterile or bacteriostatic water added slowly against the inner wall of the vial. Allow the cake to dissolve without agitation, swirling gently if required. Do not vortex or shake.

Stability

Lyophilised at −20 °C, desiccated and protected from light, stable for at least 24 months. Reconstituted and held at 2–8 °C, use within 28 days. Avoid repeated freeze-thaw.

Notes

  • Bring the vial to room temperature before opening so atmospheric moisture does not condense onto cold powder.
  • Record the reconstitution date on the vial; concentration drift between runs is usually evaporation, not synthesis variance.
  • Aliquot at first reconstitution where the working programme needs more than a few withdrawals.

Why will it not dissolve in water?
The central leucine-rich stretch is strongly hydrophobic. Prepare a DMSO stock first, then dilute into aqueous buffer immediately before use. Going straight into water usually gives a suspension, and a concentration that reads low on assay.
Which analogue do you supply?
Wild-type human humanin, unmodified. The S14G analogue is a different, more potent variant and is quoted separately on request.
Does it aggregate in storage?
Not as the lyophilised powder. Aggregation is a solution-phase problem, which is why we recommend preparing fresh rather than holding aqueous stocks.
