About Epitalon

Epitalon is a four-residue peptide, Ala-Glu-Asp-Gly, developed as a synthetic counterpart to the pineal extract epithalamin. It is one of the shortest sequences in regular research use, and its small size and high polarity make it straightforward to handle but unusual to assay — it elutes very early on standard reversed-phase gradients and often needs a modified method.

The research interest centres on reported effects on telomerase expression in somatic cell cultures and on circadian and pineal regulation. Much of the primary literature is Russian-language and older, and secondary sources frequently overstate it, so the compound is best approached with the original methodology in hand.

Reported induction of TERT expression in somatic cell culture is the most-cited line of work on the compound.

Circadian and pineal regulation

Derived from pineal extract research, the peptide appears in work on melatonin rhythm and pineal function.

The compound is a standard example in the broader literature on very short regulatory peptides and their proposed mechanism of action.

Handling

Reconstitution and stability

Reconstitution

Reconstitute with sterile or bacteriostatic water added slowly against the inner wall of the vial. Allow the cake to dissolve without agitation, swirling gently if required. Do not vortex or shake.

Stability

Lyophilised at −20 °C, desiccated and protected from light, stable for at least 24 months. Reconstituted and held at 2–8 °C, use within 28 days. Avoid repeated freeze-thaw.

Notes

  • Bring the vial to room temperature before opening so atmospheric moisture does not condense onto cold powder.
  • Record the reconstitution date on the vial; concentration drift between runs is usually evaporation, not synthesis variance.
  • Aliquot at first reconstitution where the working programme needs more than a few withdrawals.

Why does it elute so early on HPLC?
It is four residues, highly polar and carries two acidic side chains. On a standard C18 gradient it can co-elute with the void. We use a modified method with a shallow initial gradient; if you are re-assaying, expect to adjust yours.
Is Epitalon the same as Epithalon?
Yes. Both spellings refer to the same AEDG tetrapeptide. Epithalamin is the pineal extract from which it was derived, and is a different, undefined preparation.
How strong is the evidence base?
Much of the primary literature is older and Russian-language, and secondary sources routinely overstate it. We supply the material to specification and leave the interpretation of the literature to the researcher.
