Overview

About 5-Amino-1MQ

5-Amino-1MQ is a small-molecule quaternary quinolinium that inhibits nicotinamide N-methyltransferase. It is not a peptide, and it is listed in the metabolic group because NNMT sits at the junction of methylation capacity and NAD+ salvage — the enzyme consumes both nicotinamide and the methyl donor SAM.

That dual consumption is what makes the inhibitor useful. Blocking NNMT raises intracellular nicotinamide available for the NAD+ salvage pathway while sparing SAM, so the compound is used to probe both arms in the same preparation rather than manipulating each separately.

In short

  • Cell-permeable NNMT inhibitor, supplied as the iodide salt
  • Not a peptide — assayed by a small-molecule HPLC method
  • Links methylation capacity to NAD+ salvage in a single intervention
  • Released at ≥ 98.0% area-percent by HPLC

Where it is used

Research applications

NAD+ salvage

Inhibiting NNMT spares nicotinamide for salvage to NAD+, making the compound a tool for raising cellular NAD+ without exogenous precursor loading.

Adipocyte metabolism

NNMT is highly expressed in adipose tissue, and inhibition is studied for its effects on adipocyte energy expenditure and differentiation.

One-carbon metabolism

Because NNMT consumes SAM, inhibition changes methyl donor availability, connecting the compound to broader methylation research.

Handling

Reconstitution and stability

Reconstitution

Reconstitute with sterile or bacteriostatic water added slowly against the inner wall of the vial. Allow the cake to dissolve without agitation, swirling gently if required. Do not vortex or shake.

Stability

Lyophilised at −20 °C, desiccated and protected from light, stable for at least 24 months. Reconstituted and held at 2–8 °C, use within 28 days. Avoid repeated freeze-thaw.

Notes

  • Bring the vial to room temperature before opening so atmospheric moisture does not condense onto cold powder.
  • Record the reconstitution date on the vial; concentration drift between runs is usually evaporation, not synthesis variance.
  • Aliquot at first reconstitution where the working programme needs more than a few withdrawals.

Questions

5-Amino-1MQ — asked and answered

Something not covered here? Ask us directly — technical questions reach someone who runs the assays.

Why is there no CAS number listed?
Public registry assignment for this salt is inconsistent across sources, and we would rather leave the field blank than publish a number we cannot stand behind. The batch certificate carries the full analytical identity.
Is this a peptide?
No. It is a small-molecule quaternary ammonium compound. It appears in this catalogue because the research use sits alongside the metabolic peptides.
Which solvent should be used?
Water for aqueous assays, DMSO where higher stock concentrations are needed. Confirm final DMSO tolerance in your cell system.
