Targets and pathways
Overview
TB-500 is supplied as thymosin beta-4, a 43-residue peptide whose primary biochemical function is sequestering monomeric G-actin. By holding actin monomers out of the polymerising pool it changes the dynamics of cytoskeletal assembly, which is the mechanism underlying its effects on cell migration.
A note on naming: TB-500 is often described as a fragment of thymosin beta-4 rather than the full peptide, and the two are used interchangeably in the trade. We supply and certify the full 43-residue sequence, stated explicitly on the certificate, so there is no ambiguity about what is in the vial.
In short
Where it is used
The actin-sequestering function makes it a direct tool for perturbing polymerisation without disrupting the cytoskeleton wholesale.
Migration and wound-closure assays are the most common application, following from the cytoskeletal mechanism.
A substantial literature exists in epithelial repair models, particularly ocular surface work.
Handling
Reconstitute with sterile or bacteriostatic water added slowly against the inner wall of the vial. Allow the cake to dissolve without agitation, swirling gently if required. Do not vortex or shake.
Lyophilised at −20 °C, desiccated and protected from light, stable for at least 24 months. Reconstituted and held at 2–8 °C, use within 28 days. Avoid repeated freeze-thaw.
Laboratory reference
Enter the fill weight on the vial and the diluent volume you intend to add. The calculator returns the resulting solution concentration and the volume that contains a given mass.
Figures are mass-to-volume arithmetic for laboratory preparation of TB-500 and take no account of net peptide content, which is stated on the batch certificate and should be applied to the fill weight before calculating. Nothing here constitutes dosing, administration or clinical guidance of any kind.
