Targets and pathways
Overview
This is GHRH(1-29) with four substitutions — D-Ala at position 2, Gln at 8, Ala at 15 and Leu at 27 — each addressing a specific degradation or aggregation liability in the native sequence. The D-Ala2 substitution is the important one: it blocks DPP-4 cleavage, which otherwise clears native GHRH within minutes.
The 'no DAC' designation matters. CJC-1295 with drug affinity complex carries a maleimide group that binds covalently to serum albumin, extending half-life to days. Without it, the analogue is short-acting, and the two forms behave completely differently in a preparation. They are separate compounds and are frequently confused in secondary sources.
In short
Where it is used
The short duration of action makes this the analogue of choice where pulsatile GHRHR stimulation is being modelled.
Comparing short- and long-acting analogues is how desensitisation kinetics at GHRHR are characterised.
Frequently paired with a ghrelin receptor agonist to study the two secretagogue pathways acting together.
Handling
Reconstitute with sterile or bacteriostatic water added slowly against the inner wall of the vial. Allow the cake to dissolve without agitation, swirling gently if required. Do not vortex or shake.
Lyophilised at −20 °C, desiccated and protected from light, stable for at least 24 months. Reconstituted and held at 2–8 °C, use within 28 days. Avoid repeated freeze-thaw.
Laboratory reference
Enter the fill weight on the vial and the diluent volume you intend to add. The calculator returns the resulting solution concentration and the volume that contains a given mass.
Figures are mass-to-volume arithmetic for laboratory preparation of CJC-1295 (no DAC) and take no account of net peptide content, which is stated on the batch certificate and should be applied to the fill weight before calculating. Nothing here constitutes dosing, administration or clinical guidance of any kind.
